Description

Nanoparticles (NP) are finding more and more their way to clinical applications: Microsensors are used for diagnostic purposes, magnetic NP are applied for cancer treatment and drugs coupled to NP are used to enhance the effect of medications. A possible side effect of NP is teh generation of autoimmune diseases mediated by macrophages. To investigate the activation of cells of the immune system by NP, NP coated surfaces of NP coated with TCM the cell lines from myeloic origin will be used. To investigate the interaction of leucocytes with NP and NP coated surfaces several bioassays will bei applied e.g. Enzyme-linked immuno-sorbent assay (ELISA) will be used to measure the secretion of cytokines. Antibodies necessary for this essay are well tested and commercially available. Expression of surface structures (MHC-1) will be detected by fluorescence labelled specific antibodies directed against MHC-1 proteins and visualised by immunofluorescence microscopy.
